Cultivation of Anerobic Bacteria
The main principle of cultivation anerobic bacteria is to reduse the oxygen cantaint of culture media and remove any oxygen which is already paresent the system in the medium
oxygen free nitrogen is bubbled through the medium to keep it anerobic
The medium is then dispensed into tubes which are being flrshed with oxygen free nitrogen and are strelized by autoclaving
A reducing agent cystein is added to further lower
(1) Pre- Redused media
For preparing a pre- reduced media the calcture media is bailed for several minute to derive of most of the oxygen contentoxygen free nitrogen is bubbled through the medium to keep it anerobic
The medium is then dispensed into tubes which are being flrshed with oxygen free nitrogen and are strelized by autoclaving

(2) Anaerobic chamber
This refer to a plastic anerobic glove box that contains an atmosphere of H2 , CO2 & N2
culture media are placed with in the chamber by means of an air lock which can be evacuated by reaction with hydrogen forming water this reaction is aided by a paladium catalyst
after being renderd oxygen free the media are inocculated with in the chamber are innoculated ( by means of the glove part ) and incubated with in the chamber
(3)Anaerobic Jar
It is a heavy walled jar with a gas tite seal whith in which tubes plate or other containrs to be incubated are pllaced along with H2 & Co2 generating system
After the jar is shield oxygen present in the atmosphere inside the jar & dissolved in the calture medium is gradually used up through reaction with the hydrogen in the presence of cataltst
The air in the jar replaced with a mi2of H2 & Co2 thus leading to anoxic condition
